Live-Cell Target Engagement

ICE Bioscience offers a comprehensive range of NanoBRET Target Engagement (TE) Assays. These assays provide real-time, quantitative data on the interaction between a test compound and an expressed target protein within live cells, supporting drug discovery and development.

NanoBRET TE technology uses bioluminescence resonance energy transfer to measure proximity between a NanoLuc luciferase-tagged target protein and a fluorescently labelled tracer that competes with test compounds. This enables quantitative assessment of drug-target engagement in a cellular context that is relevant to physiological conditions.

Induced Proximity Applications
Cellular Target Engagement for PROTACs, Molecular Glues, and RIPTACs

NanoBRET TE assays can be applied to induced-proximity programs to measure whether PROTACs, molecular glues, RIPTACs, or related test compounds engage their target proteins in live cells.

For RIPTAC programs, NanoBRET can help assess engagement of the selected target protein in a cellular context and support the interpretation of target-proximal activity.

These cellular target-engagement data can complement downstream degradation, ternary-complex, signaling, or functional assays by helping distinguish insufficient target engagement from later-stage biological effects.
