DUB enzymatic activity assays use fluorescence intensity (FI) measurements with Rhodamine 110. In this format, Rhodamine 110 is conjugated to ubiquitin. When a DUB cleaves the ubiquitin–Rhodamine 110 conjugate, fluorescent Rhodamine 110 is released. The resulting increase in fluorescence intensity is measured using a high-sensitivity microplate reader and reflects DUB enzymatic activity.
